Abstract   FAM(5-fluorouracil, adriamycin, mitomycin-C) combination chemotheapy regimen is widely used in stomach cancer patients. The in vitro effect of therapeutic concentrations of FAM on the phagocytosis of Staphylococcus aureus by human polymorphonuclear leukocytes(PMNL) was assessed with flow cytometry. PMNL were isolated from whole blood of eleven healthy controls and eleven patients with untreated gastric cancer by density gradient centrifugation. Cells were incubated with 5-fluouracil 51.8 ug/ml, adriamycin 2.6ug/ml, and mitomycin-C 0.87 pg/ml. The phagocytic activity of PMNL was compared at times 0, 1, and 4 hours after incuba- tion. 1) The phagocytic activity was significantly lower in patients with gastric cancer compared with controls at baseline(p<0.05). 2) The phagocytic activity decreased in both controls and patients at 1 hours of incubation (p<0.05). The phagocytic activity at 1 hours was significantly lower in patients than in controls. 3) The phagocytic activity showed tendency to increase at 4 hours as compared to that of 1 hours in both controls and patients. In conclusion, the baseline phagocytic activity was significantly lower in patients with gastric cancer and FAM reduces phagocytic activity of PMNL at 1 hours in both controls and patients. The phagocytic activity was significantly lower in the patients than in controls at 1 and 4hours after treatment with FAM.
